The cholinergic neurons of the upper brain stem are found within the nucleus pedunculopontinus Ch5 ; and laterodorsal tegmental nucleus Ch6 ; [14, 23]. Three of these sectors were shown to be involved in learning and memory functions: Ch1, Ch2 and Ch4. Lesions to these areas produced severe mnemonic deficits in rodents [24, 25]. The topographical organization of the cholinergic projections to the cortex is limited. Neurons located in the anterior part of the Ch4 region primarily innervate lateral frontal and parietal regions, while neurons found in more posterior areas of Ch4 project to more temporal areas [26]. In general, the anatomical organization of cortical cholinergic afferents suggests a widespread, undifferentiated innervation of cortical areas by basal forebrain cholinergic neurons, rather than activation of a specific cortical region system. Specifically, cortical cholinergic inputs mediate the subjects' abilities to detect and select stimuli and associations for extended processing and to allocate the appropriate processing resources to these functions. Lesions to the primary motor or the prefrontal cortex produced analogous motor and behavioural alterations in rodents and primates. Among the impaired functions after lesions to these areas, primates and rodents present difficulty in shifting responses, poor performance in spatial learning tasks, reduced social interaction, and impaired behavioural habituation [27, 28]. 1.5 Animal Models of Neuropsychopathology Animal models have played a major role in efforts to determine the behavioural and neuronal mechanisms underlying drug effects and have empirically served to develop and test many neuropsychopathological theories. An animal model can be described as an experimental preparation developed in one species for the purpose of studying a phenomena occurring in another. The general argument for the use of animals in behavioural research is that such models allow the testing of specific hypothesis under controlled conditions using methods that are considered either impossible or unethical to use in humans. Muscle and ocular symptoms occur generally two to eight weeks after the gastrointestinal symptoms. Cardiac and neurologic complications may appear in the third to sixth week and, in most severe cases, death due to myocardial failure may occur. Week. The tonographic data and aqueous humor ascorbate ratios indicate that demeclocycline decreases IOP by suppressing aqueous humor formation. In addition, the increase in aqueous humor protein is also consistent with the decrease in aqueous humor formation. No untoward effects such as inflammation were observed during these studies, and histologic examination of three eyes at the time of maximum IOP drop revealed no evident pathology by light microscopy. A similar ocular hypotensive effect occurs in cats following intravitreal demeclocycline. The nature of the hypotensive response and the properties of these drugs may explain why their pronounced effects on intraocular pressure have been so long unappreciated. Available antiglaucoma agents act within a few hours, and the onset of the tetracycline-induced ocular hypotension is delayed long beyond the time course of most studies of aqueous humor dynamics. In addition, the tetracyclines as currently formulated penetrate poorly into the eye after topical administration.1314 In the current studies, we specifically used intravitreal injections to bypass the poor ocular penetration of these agents. The biochemical mechanism by which tetracyclines decrease intraocular pressure is unclear. The unilaterality of the effect argues against a systemic action, and the ineffectiveness of systemic drug administration in lowering IOP indicates that intraocular drug deposition is critical. While demeclocycline inhibits the acute rise in IOP induced by desmopressin, 4 there is no compelling evidence for tonic vasopressin activity on the ciliary epithelium, and the long delay required for tetracyclines to lower IOP suggests an alternative mechanism. Some of the neuropeptides in the eye cause prolonged physiological effects in other systems, 1 and tetracyclines may be interacting with an as yet undiscovered neuroendocrine mechanism in the eye. In this regard, the delayed onset of their effect is consistent even with retrograde axoplasmic transport to peripheral ganglia innervat.
