A frequent topic of discussion among women, osteoporosis also affects men, though in lower numbers. P6 An MPV Outbreak Likely Originating from Sentinels, Determination of Prevalence and Sensitivity of Serology, Mesenteric Lymph Node and Fecal PCR S Maguire1, * , G Marks2, R Jackson1 Bristol Myers Squibb, Pharmaceutical Research Institute, 1Hopewell, NJ, 2Wallingford, CT Our facility health monitoring program first detected mouse parvovirus MPV ; by serologic immunofluorescent assay IFA ; and mesenteric lymph node polymerase chain reaction MLN ; after terminal bleeds and collection of mesenteric lymph nodes of Hsd: ICR CD-1 ; sentinels in a room housing the breeding program which produced the facility sentinels . For purposes of this report, the gold standard of MPV testing was a combination of IFA and MLN; mice were considered MPV positive with either a positive IFA or MLN, and were considered MPV negative with a negative IFA and MLN . Further IFA and MLN testing revealed MPV-positive sentinels in three rooms housing research animals as well as confirming MPV in the sentinel breeding program . MPV testing by polymerase chain reaction of fecal pellets collected directly from mice at necropsy and from cages was instituted to assess the sensitivity of fecal PCR . MPVpositive and -negative cages from the sentinel breeding program were identified and placed in a flexible film isolator to study MPV transmission within the isolator over time . Additional terminal testing of sentinel, sentinel breeding program, and research mice was completed throughout the facility . Over 7 weeks, one of seven cages became MPV positive in the isolator . Of the 214 research mice from the sentinel positive rooms, all tested MPV negative, while 14 of the 70 mice from the sentinel breeding program were MPV positive, localizing the likely source of the outbreak to the sentinel breeding program . In testing of 138 ICR sentinel and sentinel breeding program mice at least 6 weeks old, MLN had the greatest sensitivity for detecting MPV 93% ; , followed by IFA 68% ; , direct fecal PCR 10% ; and cage fecal PCR 5% ; , and the prevalence of MPV within cages containing at least one positive animal was 60 and stavudine.

Evidence that resistance of T. brucei to organo-arsenicals is associated with selective uptake rather than site of action, was reported by Williamson 1959 ; . He showed that melaminophenyl arsenicals and diamidines are selectively antagonised by melamine. Melarsoprol resistance was first linked to the absence of the P2 aminopurine transporter by Carter and Fairlamb 1993 ; , who systematically tested a large number of metabolites for their ability to block melarsen-oxide-induced lysis in vitro. They found that, of the compounds tested only adenosine and adenine displayed this ability, and went on to characterise two T. b. brucei adenosine transporters, P1 and P2. Only P2 was sensitive to adenine and the P2 transport activity was lacking in a sodiummelarsen-adapted cloned line of T. b. brucei, cRU15 Carter and Fairlamb, 1993 ; , which was highly resistant to all melaminophenyl arsenicals in vivo Fairlamb et al., 1992 ; . In the field, no cross resistance between isometamidium and diminazene is observed which would have been the case if the resistance mechanisms were the same for the two compounds. Between homidium and isometamidium cross resistance is actually observed allowing the hypothesis of a common mechanism. Our observations led us to the conclusion that all diamidines do not share the same uptake mechanisms and that the strategies to become resistant to those compounds are not the same. Melarsoprol and diminazene on one hand and isometamidium and homidium on the other hand seem to loose toxicity for the drug resistant trypanosomes by distinct pathways. Some particularities of the uptake of diamidine compounds in T. brucei and in T.congolense will be presented and discussed.

Neuronal nitric oxide synthase nNOS ; -derived nitric oxide NO ; plays an important role in kidney homeostasis by regulating sodium and water excretion and renal medullary blood flow. In the heart, it enhances -adrenergic-stimulated contractility by increasing SR Ca + release. Despite the discovery of an endogenous protein inhibitor of nNOS PIN ; , little is known about its distribution and regulation in the kidney and heart of spontaneously hypertensive rats SHR ; . Accordingly, our objective was to study the expression of PIN and nNOS in heart and kidneys obtained from male SHR and the normotensive control Wistar rats 14-16 weeks-old ; . Our results show that in kidneys from SHR, protein expression of PIN was significantly decreased average 58%, p 0.01 ; and nNOS was significantly increased average 72%, p 0.05 ; . In renal cortex the gene expression of PIN was significantly increased average 33.31%, p 0.05 ; and no difference was found in nNOS. In renal medulla no difference in the gene expression of PIN and nNOS was observed compared to control rats. On the other hand, in the heart of SHR, no difference in the protein expression of PIN was observed compared to control rats. The altered expression of both PIN and nNOS in SHR kidneys may contribute to hypertensive state. However, the physiological significance of these alterations still remains to be elucidated.
